Elisa試劑盒實驗原理：用純化的抗體包被微孔板，制成固相載體，往包被抗LEP抗體的微孔中依次加入標本或標準品、*化的抗LEP抗體、HRP標記的親和素，經過*洗滌后用底物TMB顯色。TMB在過氧化物酶的催化下轉化成藍色，并在酸的作用下轉化成zui終的黃色。顏色的深淺和樣品中的LEP呈正相關。用酶標儀在450nm波長下測定吸光度（OD值），計算樣品濃度

樣品類型（SAMPLE TYPES）：標本必須為液體，不含沉淀。包括血清、血漿、尿液、胸腹水、腦脊液、細胞培養上清、組織勻漿等。1ml的全血可得到0.5ml的血清或血漿。每個標本量收集體積＝100ul×檢測種類。取材前須向銷售人員索要說明書。 

樣品采集：收集標本前必須清楚要檢測的成份是否足夠穩定。對收集后當天進行檢測的標本，儲存在4℃備用，如有特殊原因需要周期收集標本，將標本及時分裝后放在-20℃或-70℃條件下保存。避免反復凍融。標本2-8℃可保存48小時，-20℃可保存1個月。-70度可保存6個月。部分激素類標本需添加*。
