elisa實驗目的
     酶聯免疫吸附測定（enzyme-linked immunosorbent assay 簡稱ELISA）是在免疫酶技術（immunoenzymatic techniques）的基礎上發展起來的一種新型的免疫測定技術,ELISA過程包括抗原（抗體）吸附在固相載體上稱為包被，加待測抗體（抗原）, 再加相應酶標記抗體（抗原）,生成抗原（抗體）--待測抗體（抗原）--酶標記抗體的復合物，再與該酶的底物反應生成有色產物。借助分光光度計的光吸收計算抗體（抗原）的量。待測抗體（抗原）的定量與有色產生成正比。

elisa實驗原理
    用于免疫酶技術的酶有很多，如過氧化物酶，堿性磷酸酯酶，β－Ｄ－半乳糖苷酶，葡萄糖氧化酶，碳酸酐酶，乙酰*酯酶，6－磷酸葡萄糖脫氧酶等。常用于ELISA法的酶有辣根過氧化物酶，堿性磷酸酯酶等，其中尤以辣根過氧化物酶為多。由于酶摧化的是氧化還原反應，在呈色后須立刻測定，否則空氣中的氧化作用使顏色加深，無法準確地定量。
