間接法是檢測(cè)抗體常用的方法。其原理為利用酶標(biāo)記的抗抗體（抗*抗體）以檢測(cè)與固相抗原結(jié)合的受檢抗體，故稱為間接法（見圖2-3）。操作步驟如下：
1）將特異性抗原與固相載體聯(lián)結(jié)，形成固相抗原。洗滌除去未結(jié)合的抗原及雜質(zhì)。
2）加稀釋的受檢血清，保溫反應(yīng)。血清中的特異抗體與固相抗原結(jié)合，形成固相抗原抗體復(fù)合物。經(jīng)洗滌后，固相載體上只留下特異性抗體，血清中的其他成份在洗滌過程中被洗去。
3）加酶標(biāo)抗抗體。可用酶標(biāo)抗人Ig以檢測(cè)總抗體，但一般多用酶標(biāo)抗人IgG檢測(cè)IgG抗體。固相免疫復(fù)合物中的抗體與酶標(biāo)抗體抗體結(jié)合，從而間接地標(biāo)記上酶。洗滌后，固相載體上的酶量與標(biāo)本中受檢抗體的量正相關(guān)。
