Anti-Runx3抗體,Runx3抗體產(chǎn)品介紹：RUNX3 binds to the core site of murine Leukemia virus, the core sequences in the enhancer of the polyomavirus, and also to the enhancers of the T-cell receptor genes. May be involved in the control of cellular proliferation and/or differentiation (By similarity). Heterodimer of an alpha and a beta subunit. The alpha subunit binds DNA as a monomer and through the Runt domain.DNA-binding is increased by heterodimerization. Interacts with TLE1 and SUV39H1, Subcellular location in Nucleus.

英文別名：This gene encodes a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex that binds to the core DNA sequence 5'-PYGPYGGT-3' found in a number of enhancers and promoters, and can either activate or suppress transcription. It also interacts with other transcription factors. It functions as a tumor suppressor, and the gene is frequently deleted or transcriptionally silenced in cancer. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

設(shè)置對(duì)照
其目的在于證明和肯定陽(yáng)性結(jié)果的特異性，排除非特異性疑問(wèn)。主要是針對(duì)*抗體對(duì)照，常用的對(duì)照方法包括：1. 陽(yáng)性對(duì)照；2. 陰性對(duì)照；3. 阻斷實(shí)驗(yàn)； 4.替代對(duì)照；5. 空白對(duì)照；6. 自身對(duì)照；7. 吸收實(shí)驗(yàn).
1. 陽(yáng)性對(duì)照用已知抗原陽(yáng)性的切片作對(duì)照與待檢標(biāo)本同時(shí)進(jìn)行免疫組織化學(xué)染色，對(duì)照切片應(yīng)呈陽(yáng)性結(jié)果，成為陽(yáng)性對(duì)照。證明全過(guò)程均符合要求，尤其當(dāng)待檢標(biāo)本呈現(xiàn)陰性結(jié)果時(shí)，陽(yáng)性對(duì)照尤為重要。
2.陰性對(duì)照用確證不含已知抗原的標(biāo)本作對(duì)照，應(yīng)呈陰性結(jié)果，成為陰性對(duì)照，是陰性對(duì)照的一種。其實(shí)空白、替代、吸收和抑制都屬于陰性對(duì)照。當(dāng)待檢標(biāo)本呈陽(yáng)性結(jié)果時(shí)，陰性對(duì)照就更加重要，用以排除假陽(yáng)性。
顯色觀察
免疫酶染色應(yīng)注意控制：
1. 呈色質(zhì)濃度和反應(yīng)時(shí)間。增加成色質(zhì)的量和（或）增加底物反應(yīng)時(shí)間，可增加反應(yīng)產(chǎn)物強(qiáng)度；著色太深可減少反應(yīng)時(shí)間。
2. 過(guò)氧化物酶顯色時(shí)，H2O2濃度較大將使顯色反應(yīng)過(guò)快而至背景加深；過(guò)量H2O2可能抑制酶的活性。
