Anti-CD44抗體,CD44抗體染色失敗的幾種原因

1.所染的全部切片均為陰性結(jié)果，包括陽(yáng)性對(duì)照在內(nèi)，全部呈陰性反應(yīng)，原因可能是：（1）染色未按嚴(yán)格操作步驟進(jìn)行；（2）漏加一種抗體，或抗體失效；（3）緩沖液內(nèi)含*，抑制了酶的活性；（4）底物中所加H2O2量少或失效；復(fù)染或脫水劑使用不當(dāng)。
2.所有切片均呈弱陽(yáng)性反應(yīng)：（1）切片在染色過(guò)程中抗體過(guò)濃，或干燥了；（2）緩沖液配制中未加氯化鈉或pH不準(zhǔn)確，洗滌不*；（3）使用已變色的呈色底物溶液，或呈色反應(yīng)時(shí)間過(guò)長(zhǎng)；（4）抗體溫育的時(shí)間過(guò)長(zhǎng)；（5）H2O2濃度過(guò)高，呈色速度過(guò)快；（6）粘附劑太厚。
3.所有切片背景過(guò)深：（1）未用酶消化處理切片；（2）切片或涂片過(guò)厚；（3）漂洗不夠；（4）底物呈色反應(yīng)過(guò)久；（5）蛋白質(zhì)封閉不夠或所用血清溶血；（6）使用全血清抗體稀釋不夠。

Anti-CD44抗體,CD44抗體產(chǎn)品介紹：CD44 is the receptor for hyaluronic acid (HA). It mediates cell to cell and cell matrix interactions through its affinity for HA. Adhesion with HA plays an important role in cell migration, tumor growth and progression. Also involved in lymphocyte activation, recirculation and homing, and in hematopoiesis. High protein heterogeneity due to several alternative splicing and post translational modification events. CD44R represents a heterogeneous group of CD44 variant isoforms expressed constitutively in epithelial cells and monocyte lineage cells and can be upregulated in activated leukocytes. CD44R, and in particular CD44v6 containing isoforms, may be involved in leukocyte attachment to and rolling on endothelial cells, homing to peripheral lymphoid organs and to sites of inflammation. CD44v6 containing isoforms may be involved in tumor metastasis.

The protein encoded by this gene is a cell-surface glycoprotein involved in cell-cell interactions, cell adhesion and migration. It is a receptor for hyaluronic acid (HA) and can also interact with other ligands, such as osteopontin, collagens, and matrix metalloproteinases (MMPs). This protein participates in a wide variety of cellular functions including lymphocyte activation, recirculation and homing, hematopoiesis, and tumor metastasis. Transcripts for this gene undergo complex alternative splicing that results in many functionally distinct isoforms, however, the full length nature of some of these variants has not been determined. Alternative splicing is the basis for the structural and functional diversity of this protein, and may be related to tumor metastasis.
