保存條件: Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
Anti-TRAP80抗體,MED17抗體產品介紹：In mammalian cells, transcription is regulated in part by high molecular weight coactivating complexes that mediate signals between transcriptional activators and RNA polymerase (1). These complexes include CRSP (for cofactor required for Sp1 activation), which is required, in conjunction with TAFIIs, for transcriptional activation by Sp1 (2). CRSP is ubiquitously expressed in various tissues and functions as a multimeric complex that consists of nine distinct subunits (3). Several members of the CRSP family share sequence similarity with multiple components of the yeast transcriptional mediator proteins, including CRSP150, which is related to yeast Rgr1, and CRSP70, which is similar to the elongation factor TFIIS (4). CRSP77 and CRSP150 are also related to proteins within the putative murine mediator complex, while CRSP130 and CRSP34 are largely unrelated to either murine or yeast proteins (2,5). CRSP subunits also associate with larger multimeric coactivaor complexes, including ARC/DRI, which binds directly to SREBP and nuclear hormone receptors to facilitate transcription, and with NAT, a polymerase II-interacting complex that represses activated transcription (6,7).
操作流程
1 脫蠟和水化：脫蠟前應將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘。 a 組織芯片置于二甲苯中浸泡10分鐘,更換二甲苯后在浸泡10分鐘
b 無水乙醇中浸泡五分鐘
c 95%乙醇中浸泡五分鐘
d 75%乙醇中浸泡五分鐘
2 抗原修復：用于福爾馬林固定的石蠟包埋組織芯片：
A 抗原熱修復
a 高壓熱修復 在沸水中加入EDTA（ph8.0）或0.01m*緩沖溶液（ph6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色加上，緩慢加壓，是玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。
b 沸熱修復 電爐或水浴鍋加熱0.01*緩沖液（ph6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
C 微波爐加熱 在微波爐里加熱0.01*緩沖液（ph6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復1-2次。適用的抗原：
Bcl-2.Bax.AR.PR.C-fos.x-jum.z-kit.c-myc.E-cadherin.
ChromograninA.Cyclin.ER.Heatshock.Protein.HPV.Ki-67.MDMZ.P53.P34.P15.P-glycoprotein.PKC.PCNA.ras.Rb.等
B 酶消化方法
常用0.1%*和0.4%*液。*使用前預熱37℃，消化時間約為5-30分鐘。適用于被固定遮蔽的抗原：包括 Collagen.GFAP.Complement.Cytokeratin.C-erB-2.LCA.LN.等
3 免疫組化染色
SP法
（1） 脫蠟、水化 （2）PBS洗2-3次各5分鐘 （3）3%H2O2（80%甲醇）滴加在
TMA上，室溫靜置10分鐘 （4）PBS洗2-3次各5分鐘 （5）抗原修復
（6）PBS洗2-3次各5分鐘 （7）滴加正常山羊血清封閉液，室溫20分鐘，甩去多余液體（8）滴加Ι抗50μl,室溫靜置1小時或4℃過夜或37℃1小時（9）4℃過夜后需在37℃復溫45分鐘（10）PBS洗3次每次2分鐘（11）滴加Ⅱ抗45-50μl,室溫靜置或37℃1小時（12）Ⅱ抗中可加入0.05%的
tween-20.（13）PBS洗3次各5分鐘（14）DAB顯色5-10分鐘，在顯微鏡下掌握染色程度 （15）PBS或自來水沖洗10分鐘 （16）蘇木精復染2分鐘，鹽酸酒精分化（17）自來水沖洗10-15分鐘 （18）脫水、透明、封片、鏡檢。
SABC法
（1） 脫蠟、水化 （2）PBS洗2次各5分鐘 （3）用蒸餾水或PBS配制新鮮的3%H2O2，室溫封閉5-10分鐘，用蒸餾水洗3次（4）抗原修復（5） PBS洗5分鐘 （6）滴加正常山羊血清封閉液，室溫20分鐘，甩去多余液體（7）滴加Ι抗50μl,室溫靜置1小時或4℃過夜或37℃1小時（8）PBS洗3次各2分鐘（9）滴加*化Ⅱ抗，20℃-37℃ 20分鐘（10）PBS洗3次各2分鐘 （11）滴加試劑SABC 20℃-30℃ 20分鐘（12） PBS洗4次各5分鐘 （13） DAB顯色：試劑盒或自配顯色劑顯色（14）脫水、透明、封片、鏡檢。
