Anti-MC-1R抗體,黑皮質素1受體抗體產品介紹：This intronless gene encodes the receptor protein for melanocyte -stimulating hormone (MSH). The encoded protein, a seven pass transmembrane G protein coupled receptor, controls melanogenesis. Two types of melanin exist: red pheomelanin and black eumelanin. Gene mutations that lead to a loss in function are associated with increased pheomelanin production, which leads to lighter skin and hair color. Eumelanin is photoprotective but pheomelanin may contribute to UV-induced skin damage by generating free radicals upon UV radiation. Binding of MSH to its receptor activates the receptor and stimulates eumelanin synthesis. This receptor is a major determining factor in sun sensitivity and is a genetic risk factor for melanoma and non-melanoma skin cancer. Over30 variant alleles have been identified which correlate with skin and hair color, providing evidence that this gene is an important component in determining normal human pigment variation.

1、 組織取材：組織塊稱重
2、 利用液氮、研缽粉碎組織塊
3、 加入RIPA緩沖液（每克組織3ml RIPA），PMSF（每克組織30μl，10 mg/ml PMSF），
利用Polytron進一步勻漿（15,000轉/分*1分鐘）維持4℃
4、 加入PMSF（每克組織30μl，10 mg/ml PMSF），冰上孵育30分鐘
5、 移入離心管4℃，約20,000g（約15,000轉）15分鐘
6、 上清液為細胞裂解液可分裝-20℃保存
7、 進行Bradford比色法測定蛋白質濃度
8、 取相同質量的細胞裂解液（體積*蛋白質濃度），并加等體積的2×電泳加樣緩沖液
9、 沸水浴中3分鐘
10、上樣
11、電泳（濃縮膠20mA，分離膠35mA）
12、電轉膜儀轉膜（100mA 40分鐘）
13、膜用麗春紅染色，膠用考馬斯亮藍染色
14、Westernblot 試劑盒顯色
