Anti-TPOR抗體,血小板生成素受體抗體產品介紹：In 1990 an oncogene, v-mpl, was identified from the murine myeloproliferative leukemia virus that was capable of immortalizing bone marrow hematopoietic cells from different lineages. In 1992 the human homologue, named, c-mpl, was cloned. Sequence data revealed that c-mpl encoded a protein that was homologous with members of the hematopoietic receptor superfamily. Presence of anti-sense oligodeoxynucleotides of c-mpl inhibited megakaryocyte colony formation. The ligand for c-mpl, thrombopoietin, was cloned in 1994. Thrombopoietin was shown to be the major regulator of megakaryocytopoiesis and plaet formation. The protein encoded by the c-mpl gene, CD110, is a 635 amino acid transmembrane domain, with two extracellular cytokine receptor domains and two intracellular cytokine receptor box motifs . TPO-R deficient mice were severely thrombocytopenic, emphasizing the important role of CD110 and thrombopoietin in megakaryocyte and plaet formation. Upon binding of thrombopoietin CD110 is dimerized and the JAK family of non-receptor tyrosine kinases, as well as the STAT family, the MAPK family, the adaptor protein Shc and the receptors themselves become tyrosine phosphorylated.
*，要用Western Blot比較不同條件下或者不同組織中，目的蛋白表達量的相對多少，前提條件是等量的蛋白上樣，才有比較的基礎。特別是在表達量不高時，上樣量的差別就很可能影響結果的分析。所以需要內參做對比。內參即是內部參照（Internal Control），對于哺乳動物細胞表達來說一般是指由管家基因編碼表達的蛋白(Housekeeping Proteins)，它們在各組織和細胞中的表達相對恒定，在檢測蛋白的表達水平變化時常用它來做參照物。在Western Blotting 實驗中，除了需要進行蛋白抽提、蛋白定量、等量蛋白上樣電泳、轉膜、靶蛋白抗體孵育、顯色等步驟以外，還需要進行內參的檢測，以校正蛋白質定量、上樣過程中存在的實驗誤差，以保證實驗結果的準確性。（上海研盟生物科技有限公司）實驗中出現的問題及處理辦法
1、為什么帶出現拖尾現象？
原因：主要是樣品融解效果不佳或分離膠濃度過大引起的；
處理辦法：加樣前離心，選擇適當的樣品緩沖液，加適量樣品促溶劑；電泳緩沖液時間過長，重新配制;降低凝膠濃度。
2、為什么帶出現紋理現象？
原因：主要是樣品不溶性顆粒引起的；
處理辦法：加樣前離心，加適量樣品促溶劑。
3、為什么電泳的條帶很粗？
原因：電泳中條帶很粗是常見的事，主要是未濃縮好的原因；
處理辦法：適當增加濃縮膠的長度，保證濃縮膠貯液的pH正確（6.7），適當降低電壓。
